Bacterial Colonization and Proliferation in Furcal Perforations Repaired by Different Materials: A Confocal Laser Scanning Microscopy Study

چکیده
Following furcal perforation, bacteria may colonize the defect and cause inflammation periodontal destruction. This study used confocal laser scanning microscopy (CLSM) to evaluate Enterococcus faecalis colonization proliferation in perforations repaired with different materials. Furcal created 55 extracted human mandibular molars were using either MTA-Angelus, Endocem, or Biodentine coronally subjected E. suspension for 21 days. The specimens then stained a LIVE/DEAD Viability Kit visualized by CLSM. minimum maximum depths of bacterial penetration into dentinal tubules 159 1790 μM, respectively, mean 713 μM. There significantly more live than dead inside (p = 0.0023) all groups, three repair materials exhibited similarly sized area 0.083). However, there significant differences numbers at circumference perforation 0.0041), higher ratio MTA-Angelus group 0.001). repair, interface between material dentin penetrate through tubules. type has effect on viability colonizing bacteria.

Laser Scanning Confocal Microscopy
Laser scanning confocal microscopy has become an invaluable tool for a wide range of investigations in the biological and medical sciences for imaging thin optical sections in living and fixed specimens ranging in thickness up to 100 micrometers. Modern instruments are equipped with 3-5 laser systems controlled by high-speed acousto-optic tunable filters (AOTFs), which allow very precise regula...

Many technological advancements of the past decade have contributed to improvements in the photon efficiency of the confocal laser scanning microscope (CLSM). The resolution of images from the new generation of CLSMs is approaching that achieved by the microscope itself because of continued development in digital imaging methods, laser technology and the availability of brighter and more photos...
متن کاملScanning and Image Reconstruction Techniques in Confocal Laser Scanning Microscopy
Taking advantage of the laser scanning methodology, confocal microscopy has been widely applied in cutting-edge biological research for its three-dimensional, high-resolution imaging capability (Pawley 1995). The principles of confocal were put forward by Marvin Minsky in 1955 (Minsky 1988), yet the experimental demonstration was not accomplished until 20 years later by Cremer brother (Cremer, ...
متن کاملConfocal Scanning Laser Microscopy of Polymer Coatings
A new nondestructive technique, confocal scanning laser microscopy (CSLM), is described which is used to characterize the topography and morphology of polymer coatings. The topography of the coating can be determined even when the coating is completely opaque. When the coating is not completely opaque, CSLM has the distinct advantage of also being able to distinguish between the coating surface...

Optical technologies are being used increasingly to perform real-time assessment of tissue abnormalities. Many groups have demonstrated that quantitative optical spectroscopic approaches, including fluorescence spectroscopy1-4 and reflectance spectroscopy,5, 6 have the potential to deliver a highly sensitive, specific, and costeffective diagnosis in real time and without the removal of tissue. ...
